Mouse Serine Peptidase Inhibitor Kazal Type 5 ELISA Kit

A sandwich ELISA from Antibodies.com enables the in vitro quantitative determination of mouse SPINK5/LEKTI (Serine Peptidase Inhibitor Kazal Type 5) in tissue homogenates, cell lysates or other biological fluids.

The pre-coated microplate platform (12 x 8 well strips) uses a colorimetric detection method. An antibody specific for SPINK5/LEKTI is immobilised on the plate; standards and samples are added and the target binds to the wells. Following incubation and washing, a Biotinylated Anti-SPINK5/LEKTI Antibody binds the captured analyte, then HRP-Avidin conjugate is added. TMB substrate produces a blue product that turns yellow after addition of stop solution; absorbance at 450 nm in a microplate reader is proportional to analyte concentration and is read against the standard curve.

Reported sensitivity is <28 pg/ml with an assay range of 78.1–5000 pg/ml and a total assay time of 4h 30m. Recovery in undiluted serum, EDTA plasma and heparin plasma averaged 91%, 91% and 84% respectively (n=5), with ranges of 80–102%, 81–101% and 80–89%. Across dilutions of 1:2, 1:4, 1:8 and 1:16, recoveries in serum were 87–91%, 87–107%, 74–101% and 92–97%. Kit components should be stored at +4°C or -20°C as instructed and the product should not be used past its expiration date.
