A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse Serine Palmitoyltransferase in tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a 96-well microplate (12 x 8 well strips) pre-coated with antibody specific for Serine Palmitoyltransferase. Standards and samples are added to the wells, followed by a Biotinylated Anti-Serine Palmitoyltransferase Antibody and an HRP-Avidin conjugate. A TMB substrate solution visualises the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of Serine Palmitoyltransferase captured in each well; concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader against a standard curve.

The kit has a sensitivity of <0.055 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Recovery data (n=5) showed serum averaging 91% (range 80–102%), EDTA plasma averaging 91% (range 81–101%), and heparin plasma averaging 84% (range 80–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8, and 1:16 dilutions across serum, EDTA plasma, and heparin plasma samples.

Kit components include a pre-coated 96-well microplate, lyophilised standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Store kit components at +4°C or −20°C as instructed; do not use past the expiration date.
