From Antibodies.com, this Mouse SPR ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse SPR in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for SPR pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells, where SPR present in each sample is bound by the immobilized antibody. Following incubation and washing, a Biotinylated Anti-SPR Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate solution is then used to visualize the HRP enzymatic reaction by catalysis to produce a blue-coloured product that changes to yellow after addition of acidic stop solution; the density of yellow is proportional to the amount of SPR captured in each well.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.061 ng/ml. Absorbance is read at 450 nm in a microplate reader, and the quoted detection type is colorimetric. The platform is a pre-coated microplate arranged as 12 x 8 well strips, and total assay time is 4 hours 30 minutes. Manufacturer recovery data show average values of 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5), and 84% for heparin plasma (range 80–89%, n=5).
