From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay is designed for the in vitro quantitative determination of mouse S100 beta (S100 Calcium Binding Protein B) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay uses a 96-well microtiter plate pre-coated with an antibody specific for S100 beta. Standards and samples are added to the wells, where the target is captured by the immobilised antibody; following washing, a biotinylated anti-S100 beta antibody is added, then an HRP-Avidin conjugate, and the enzymatic reaction is visualised with TMB substrate solution, producing a blue product that turns yellow on addition of acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader.

Quantitative performance figures supplied by the manufacturer include a product range of 15.6–1000 pg/ml and a sensitivity of <5.9 pg/ml. Reported spike-and-recovery averages were 91% in serum (n=5, range 83%–100%), 96% in EDTA plasma (n=5, range 91%–102%) and 95% in heparin plasma (n=5, range 89%–102%). Dilution linearity across 1:2, 1:4, 1:8 and 1:16 dilutions gave recoveries of 75–104% in serum, 83–110% in EDTA plasma and 72–105% in heparin plasma. The total assay time is 4 hours 30 minutes.

Kit components comprise a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard (2 vials), Detection Solution A, Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers. Components are stored at +4°C or -20°C as instructed in the protocol, and the kit should not be used past its expiration date.
