Mouse Ribonucleotide Reductase M1 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse Ribonucleotide Reductase M1 (RRM1) in tissue homogenates or other biological fluids. The assay employs an antibody specific for RRM1 pre-coated onto a microplate, with standards and samples added so that RRM1 is captured by the immobilized antibody. Following incubation and washing, a biotinylated anti-RRM1 antibody binds the captured analyte, and an HRP-avidin conjugate is added. TMB substrate is then used to visualize the reaction; catalysis produces a blue product that turns yellow upon addition of the acidic stop solution. The intensity of yellow, proportional to the amount of RRM1 captured, is read as O.D. absorbance at 450 nm using a colorimetric microplate reader.

Reported performance characteristics include a sensitivity of <0.124 ng/ml and a measuring range of 0.312-20 ng/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. Spike-and-recovery testing in serum samples (n=5) yielded values of 80%-102% (average 91%), in EDTA plasma (n=5) 81%-100% (average 90%), and in heparin plasma (n=5) 80%-89% (average 84%). Linearity-of-dilution assessments across 1:2, 1:4, 1:8 and 1:16 dilutions gave ranges of 87-91%, 87-107%, 74-101% and 92-97% respectively for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (each n=5).
