The kit employs a pre-coated microplate, a biotinylated anti-REG3a detection antibody, and an HRP-Avidin conjugate, with signal developed using a TMB substrate and quantified by absorbance at 450 nm on a standard colorimetric microplate reader.

Reported sensitivity is 6.3 pg/ml with an assay range of 15.6-1000 pg/ml, and total assay time is approximately 4 hours 30 minutes. Spike-and-recovery averages were 91% in serum, 94% in EDTA plasma, and 94% in heparin plasma. Dilution linearity was demonstrated across dilutions of 1:2 through 1:16 in serum, EDTA plasma, and heparin plasma.
