Mouse Pyruvate Kinase, Muscle ELISA Kit

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (ELISA) is designed for the in vitro quantitative determination of mouse Pyruvate Kinase Isozyme M1 in serum, plasma or other biological fluids. An antibody specific for the target is pre-coated onto a 96-well microtiter plate and standards or test samples are added to the wells.

Captured analyte is detected using a Biotinylated Anti-Pyruvate Kinase Isozyme M1 Antibody followed by an HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the concentration is calculated from the standard curve. Total assay time is 4h 30m.

Performance characteristics include a detection range of 0.312-20 ng/ml and a sensitivity of <0.144 ng/ml. Recovery in serum averaged 91% (range 80%-102%, n=5), in EDTA plasma 90% (range 81%-100%, n=5) and in heparin plasma 84% (range 80%-89%, n=5). The kit contains a pre-coated 96-well microplate (12 x 8 well strips), lyophilised standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, with components stored at +4°C or -20°C as instructed.
