The Antibodies.com Mouse Pyruvate Dehydrogenase Alpha ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse PDHA1 in serum, plasma, tissue homogenates, cell lysates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique on a pre-coated microplate arranged as 12 x 8 well strips. An antibody specific for PDHA1 is immobilised on the plate to capture PDHA1 from standards and samples. After incubation and washing, a Biotinylated Anti-PDHA1 Antibody binds the captured PDHA1, and an HRP-Avidin conjugate is then added. TMB substrate solution is used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of an acidic stop solution. The intensity of the yellow colour is proportional to the amount of PDHA1 captured in each well, and concentration is calculated by reading absorbance at 450 nm against the standard curve.

The kit has a reported sensitivity of <0.042 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Detection is colorimetric and is measured using a microplate reader. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Reported recovery values are serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%), and heparin plasma 80%–89% (average 84%). Sample dilution linearity has been validated at 1:2, 1:4, 1:8, and 1:16 ratios for serum, EDTA plasma, and heparin plasma samples.
