Mouse Protein Z Dependent Protease Inhibitor ELISA Kit

Manufactured by Antibodies.com, this sandwich ELISA is designed for the in vitro quantitative determination of mouse ZPI (Protein Z Dependent Protease Inhibitor) in plasma samples. The assay uses a pre-coated microplate immobilised with an antibody specific for ZPI. Standards and samples are added to the wells, allowing ZPI to be captured by the coated antibody. After washing, a biotinylated anti-ZPI detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate produces a blue colour that turns yellow upon addition of the stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. The ZPI concentration is calculated by reference to a standard curve.

Sensitivity is reported as <0.41 ng/ml, with a working assay range of 0.937-60 ng/ml. Total assay time is approximately 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.

Spike-and-recovery performance has been validated across sample types (n=5 each): serum 80%-102% (average 91%), EDTA plasma 81%-100% (average 90%), and heparin plasma 80%-89% (average 84%). Linearity has been demonstrated across serial dilutions from 1:2 to 1:16. For serum, recoveries were 87-91% at 1:2, 87-107% at 1:4, 74-101% at 1:8 and 92-97% at 1:16. For EDTA plasma: 90-105%, 84-101%, 90-101% and 79-108%. For heparin plasma: 84-95%, 92-105%, 82-105% and 89-91%.
