The assay employs a 96-well microplate pre-coated with antibody specific for Calcineurin B/CNB, with standards and samples added to the wells. Following incubation and washing, a biotinylated anti-Calcineurin B/CNB antibody binds the captured target, followed by HRP-avidin conjugate. Signal is developed using TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution; the absorbance is read at 450 nm in a microplate reader and concentration is calculated from a standard curve.

The stated assay range is 0.156–10 ng/ml with a sensitivity of 0.042 ng/ml. Total assay time is 4 hours 30 minutes. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
