Mouse Protein Kinase C Delta ELISA Kit

Designed by Antibodies.com for the in vitro quantitative determination of mouse PKC delta, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is validated for use with serum, plasma, tissue homogenates and other biological fluids. The assay uses a pre-coated microtiter plate to which an antibody specific for PKC delta has been immobilised. Standards and samples are added to the wells and any PKC delta present is bound by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-PKC delta Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of PKC delta captured in each well, allowing the concentration to be calculated by reading absorbance at 450 nm in a microplate reader against a standard curve.

Validated with serum, EDTA plasma and heparin plasma samples, the assay reports a sensitivity of 0.54 ng/ml and a working range of 1.56-100 ng/ml, with a total assay time of 4h 30m. Typical recovery values across five replicates are 80-102% in serum (average 91%), 81-100% in EDTA plasma (average 90%) and 80-89% in heparin plasma (average 84%).
