Mouse Protein Kinase C Alpha ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse Protein Kinase C Alpha (PKC alpha) in tissue homogenates or other biological fluids. It employs the sandwich enzyme immunoassay technique using an antibody specific for PKC alpha that has been pre-coated onto a microplate. Standards and test samples are added to the wells, followed by a biotinylated anti-PKC alpha antibody and an HRP-avidin conjugate. After incubation and washing, TMB substrate is added; the reaction is stopped with acidic stop solution and the absorbance is read at 450 nm on a colorimetric microplate reader, with the concentration of PKC alpha calculated from the standard curve.

The quantitative assay covers a working range of 0.781–50 ng/ml with a sensitivity of <0.31 ng/ml and a total assay time of 4 hours 30 minutes. Spike-and-recovery has been validated across n=5 samples per matrix, giving ranges of 80%–102% (average 91%) for serum, 81%–100% (average 90%) for EDTA plasma, and 80%–89% (average 84%) for heparin plasma. Linearity on dilution has also been demonstrated for serum, EDTA plasma, and heparin plasma across 1:2, 1:4, 1:8, and 1:16 sample dilutions.

Kit components should be stored at +4°C or −20°C as instructed, and the kit must not be used past its expiration date. The kit is intended for use with mouse samples and is suitable for tissue homogenates and other biological fluids.
