Description

Manufactured by Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Protease Activated Receptor 2 (PAR2) in tissue homogenates, cell lysates, or other biological fluids. The assay uses a 96-well microplate pre-coated with an antibody specific for PAR2; standards and samples are added to the wells where PAR2 is captured by the immobilized antibody. Bound PAR2 is then detected using a biotinylated anti-PAR2 antibody, an HRP-avidin conjugate, and TMB substrate. The HRP-catalysed reaction produces a blue product that turns yellow upon addition of the acidic stop solution, with the intensity of yellow colour directly proportional to the amount of PAR2 captured. Concentration is calculated from the O.D. absorbance read at 450 nm.

Key performance specifications include a sensitivity of <0.053 ng/ml and an assay range of 0.156–10 ng/ml. The kit runs on a pre-coated microplate platform (12 x 8 well strips) with colourimetric detection, and the total assay time is 4 hours 30 minutes.

Reported average recovery values were 91% for serum (range 80%–102%, n=5), 90% for EDTA plasma (range 81%–100%, n=5), and 84% for heparin plasma (range 80%–89%, n=5). Linearity was assessed at dilution factors of 1:2, 1:4, 1:8, and 1:16 across serum, EDTA plasma, and heparin plasma samples.
