Mouse Prostaglandin Endoperoxide Synthase 2 ELISA Kit

From Antibodies.com, this kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse Prostaglandin Endoperoxide Synthase 2 (COX2/Cyclooxygenase 2) in serum, plasma, or other biological fluids. An antibody specific for COX2/Cyclooxygenase 2 is pre-coated onto a 96-well microtiter plate; standards and samples are added, the target is captured by the immobilised antibody, and detection uses a biotinylated anti-COX2/Cyclooxygenase 2 antibody with HRP-avidin conjugate. TMB substrate is then added and the enzymatic reaction is stopped with an acidic solution, producing a yellow colour whose intensity is proportional to the target concentration in each well. Absorbance is read at 450 nm in a microplate reader against a standard curve.

The quantitative assay delivers a sensitivity of <35 pg/ml with a measuring range of 78.1–5000 pg/ml. Detection is colourmetric on a pre-coated 12 × 8-well microplate strip platform, and the total assay time is 4 h 30 min. Validation reports the following recovery ranges (with averages): serum (n=5) 80% – 102%, average 91%; EDTA plasma (n=5) 81% – 100%, average 90%; heparin plasma (n=5) 80% – 89%, average 84%. Linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across the same sample types.

Storage conditions require kit components to be kept at +4 °C or –20 °C as instructed, and reagents must not be used past the stated expiration date.
