Mouse Podocalyxin ELISA Kit

This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse PODXL in tissue homogenates, cell lysates or other biological fluids. The assay uses a pre-coated 96-well microplate arranged as 12 x 8 well strips, with a biotinylated anti-PODXL detection antibody and HRP-avidin conjugate. Following incubation with TMB substrate, the enzymatic reaction is stopped and absorbance is read at 450 nm on a colorimetric microplate reader, with concentration calculated against the standard curve.

The quantitative assay covers a range of 0.156–10 ng/ml with a reported sensitivity of <0.041 ng/ml, and the full protocol runs in approximately 4 hours 30 minutes. Supplied components include a pre-coated 96-well microplate, lyophilized standard (2 vials), detection solution A (120 μl), detection solution B (120 µl), 30X wash buffer (20 ml), sample dilution buffer (45 ml), TMB substrate (9 ml), stop solution (6 ml) and 5 adhesive plate sealers, with storage at +4°C or –20°C as indicated on each component.

The kit is suited to cell lysates and tissue homogenates. Reported spike-and-recovery averages are 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity is provided for 1:2, 1:4, 1:8 and 1:16 sample dilutions across serum, EDTA plasma and heparin plasma (n=5).
