Antibodies.com supplies a sandwich ELISA kit for the in vitro quantitative determination of mouse PDGF AA in serum, plasma or other biological fluids. The assay uses a pre-coated microplate onto which an antibody specific for PDGF AA has been immobilised. Standards and test samples are added to the wells, followed by a biotinylated anti-PDGF AA antibody and an HRP-avidin conjugate. TMB substrate is then added; the enzymatic reaction produces a yellow product whose absorbance is read at 450 nm in a microplate reader and the concentration of PDGF AA is calculated by reference to the standard curve.

The kit reports an assay range of 31.2-2000 pg/ml and a sensitivity of <12.9 pg/ml. Recovery data from the manufacturer are given for serum (n=5, range 80-102%, average 91%), EDTA plasma (n=5, range 81-100%, average 90%) and heparin plasma (n=5, range 80-89%, average 84%). Linearity has been evaluated at sample dilutions of 1:2, 1:4, 1:8 and 1:16 for each of these sample types. Total assay time is 4 hours 30 minutes and the readout is colorimetric. Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
