Mouse Platelet Activating Factor Acetylhydrolase 2 ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (ELISA) kit from Antibodies.com is designed for the in vitro quantitative determination of mouse Platelet Activating Factor Acetylhydrolase 2 (PAFAH2) in serum, plasma, tissue homogenates or other biological fluids. The assay uses the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate. Standards and samples are added to the wells, and captured PAFAH2 is detected using a Biotinylated Anti-PAFAH2 Antibody followed by an HRP-Avidin conjugate. A TMB substrate solution visualises the reaction, producing a blue product that changes to yellow upon addition of an acidic stop solution. PAFAH2 concentration is calculated by reading the optical density at 450nm in a microplate reader against the standard curve. Sensitivity is reported as <0.062 ng/ml with an assay range of 0.156–10 ng/ml. The total assay time is 4h 30m. Kit components are stored at +4°C or –20°C as instructed.

Manufacturer recovery data (n=5) show average values of 91% in serum (range 80%–102%), 91% in EDTA plasma (range 81%–101%) and 84% in heparin plasma (range 80%–89%). Linearity data for 1:2, 1:4, 1:8 and 1:16 dilutions are: serum 87–91%, 87–107%, 74–101%, 92–97%; EDTA plasma 90–105%, 84–101%, 90–101%, 79–108%; heparin plasma 84–95%, 92–105%, 82–105%, 89–91%. The product is intended for research use only.

Required materials not supplied with the kit include a microplate reader capable of measuring absorbance at 450nm ± 10nm, a multichannel pipette reservoir or automated microplate washer, computer software capable of generating logarithmic functions, test tubes and Eppendorf tubes, multi- and single-channel pipettes with sterile tips, deionised or distilled water, absorbent paper towels, and a 37°C incubator.
