Description
From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Plasminogen Activator Inhibitor 1 (PAI1) in serum, plasma or other biological fluids. The kit employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate arranged as 12 x 8 well strips, with an antibody specific for PAI1 immobilised on the plate. Standards and test samples are added into the wells and bound PAI1 is detected with a Biotinylated Anti-PAI1 Antibody, followed by an HRP-Avidin conjugate.
The enzymatic reaction is visualised using TMB substrate solution, producing a blue-coloured product that changes to yellow after the addition of an acidic stop solution. Absorbance is read at 450 nm in a colorimetric microplate reader and the PAI1 concentration is calculated by reference to a standard curve. Sensitivity is reported as <23.7 pg/ml and the assay range as 62.5–4000 pg/ml. The total assay time is approximately 4 hours 30 minutes. Kit components are stored at +4°C or –20°C as instructed, and the kit should not be used past its expiration date.
Manufacturer-reported sample recovery averages 91% for serum (range 80–102%, n=5), 90% for EDTA plasma (range 81–100%, n=5) and 84% for heparin plasma (range 80–89%, n=5). Dilution linearity data across 1:2, 1:4, 1:8 and 1:16 sample dilutions are also provided for serum, EDTA plasma and heparin plasma.
