Mouse Pigment Epithelium Derived Factor ELISA Kit

A sandwich enzyme-linked immunosorbent assay (ELISA) from Antibodies.com, this Mouse PEDF ELISA Kit is designed for the in vitro quantitative determination of mouse Pigment Epithelium Derived Factor (PEDF) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. Detection is colourmetric on a pre-coated microplate, read at 450 nm in a microplate reader.

The quantitative sandwich format has a working range of 0.781–50 ng/ml and a sensitivity of <0.35 ng/ml, with a total assay time of 4 h 30 min. Spiked-sample recovery averaged 91% in serum (n=5, range 80%–102%), 90% in EDTA plasma (range 81%–100%) and 84% in heparin plasma (range 80%–89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 was 87–91%, 87–107%, 74–101% and 92–97% in serum (n=5); 90–105%, 84–101%, 90–101% and 79–108% in EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% in heparin plasma. Kit components are stored at +4 °C or –20 °C as instructed, and the kit should not be used past its expiration date.

Detection relies on a biotinylated anti-PEDF antibody followed by an HRP-avidin conjugate, with TMB substrate producing a blue product that turns yellow on addition of stop solution. The yellow intensity measured at 450 nm is proportional to PEDF concentration and is read against a standard curve.
