The kit employs a pre-coated 96-well microplate arranged as 12 x 8 well strips and uses a colorimetric detection method read at 450 nm in a microplate reader, with a biotinylated anti-SPLASH detection antibody and an HRP-avidin conjugate producing a TMB-based signal that is stopped with an acidic solution.

According to the manufacturer, the assay has a sensitivity of 12.4 pg/ml and a working range of 31.2–2000 pg/ml, with a total assay time of 4 h 30 min. Reported average recoveries across the tested sample types were 91% for serum, 94% for EDTA plasma and 94% for heparin plasma, and linearity was demonstrated across dilutions from 1:2 to 1:16 in each matrix.
