Mouse Phosphoenolpyruvate Carboxykinase 1, Soluble ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) kit from Antibodies.com designed for the in vitro quantitative determination of mouse phosphoenolpyruvate carboxykinase 1, soluble (PCK1/PEPC) in tissue homogenates or other biological fluids. The assay is performed on a pre-coated 96-well microplate (12 x 8 well strips) using colourmetric detection read with a standard microplate reader. The principle uses an antibody specific for PCK1/PEPC pre-coated on the plate; standards or samples are added, captured analyte is detected with a Biotinylated Anti-PCK1/PEPC Antibody, then visualised via an HRP-Avidin conjugate and TMB substrate. The reaction produces a blue product that turns yellow after addition of acidic stop solution; absorbance is read at 450nm and quantified against a standard curve.

The reported assay range is 0.156-10 ng/ml and the sensitivity is <0.058 ng/ml. Total assay time is 4h 30m. Validation data supplied include recovery in serum (n=5, range 80%-102%, average 91%), EDTA plasma (n=5, range 81%-100%, average 90%) and heparin plasma (n=5, range 80%-89%, average 84%), plus dilution linearity at 1:2, 1:4, 1:8 and 1:16 for serum (87-91%, 87-107%, 74-101%, 92-97%), EDTA plasma (90-105%, 84-101%, 90-101%, 79-108%) and heparin plasma (84-95%, 92-105%, 82-105%, 89-91%). Kit components are stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
