Mouse Peptidoglycan Recognition Protein 1 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of mouse Peptidoglycan Recognition Protein 1 (PGRP-S) in serum, plasma, tissue homogenates or other biological fluids. Reactivity is with mouse samples, and the assay is run on a pre-coated 96-well microplate.

The microplate is supplied as 12 x 8 well strips pre-coated with antibody specific for PGRP-S. Standards and samples are added to the wells and the target is captured by the immobilized antibody. After washing, a Biotinylated Anti-PGRP-S Antibody binds the captured target, and HRP-Avidin conjugate is added. TMB substrate is used to visualize the enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of stop solution. Absorbance is read at 450 nm in a microplate reader, and the PGRP-S concentration is calculated by reference to the standard curve. Total assay time is 4h 30m.

Reported sensitivity is <5.9 pg/ml and the assay range is 15.6-1000 pg/ml. In validation work, average recovery (n=5) was 91% for serum (range 80%-102%), 91% for EDTA plasma (range 81%-101%), and 84% for heparin plasma (range 80%-89%). Linearity was examined at 1:2, 1:4, 1:8, and 1:16 dilutions across the same sample types. Kit components include the pre-coated microplate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Components should be stored at +4°C or -20°C as instructed, and the kit must not be used past its expiration date.
