Description

A competitive enzyme-linked immunosorbent assay (cELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse Pancreatic Polypeptide in serum, plasma or other biological fluids. The assay targets Pancreatic Polypeptide, is reactive with mouse samples, and is listed under the Assays and Kits category.

The test uses the competitive enzyme immunoassay technique on a 96-well microtiter plate pre-coated with Pancreatic Polypeptide antigen. During incubation, Pancreatic Polypeptide in samples or standards competes with the fixed amount of immobilized antigen for binding sites on a Biotinylated Anti-Pancreatic Polypeptide Antibody. Unbound antibody is removed by washing, after which an HRP-Avidin conjugate is added to the wells. A TMB substrate solution is then used to visualise the enzymatic reaction; the catalysed product is blue and changes to yellow upon addition of acidic stop solution. The density of yellow colour is inversely proportional to the amount of Pancreatic Polypeptide present, and the concentration is calculated by reading the absorbance at 450 nm in a microplate reader against a standard curve.

Quoted performance figures from the manufacturer include a sensitivity of <8.55 pg/ml and an assay range of 31.2–2000 pg/ml, with a total assay time of approximately 2 h 30 min and detection on a colorimetric microplate reader. Reported mean recoveries are 91% in serum (n=5, range 80–102%), 90% in EDTA plasma (n=5, range 81–100%), and 84% in heparin plasma (n=5, range 80–89%). Dilution linearity is provided across 1:2, 1:4, 1:8 and 1:16 dilutions for each of the three sample types. Kit components should be stored at +4°C or -20°C as instructed and the kit should not be used past its expiration date.
