From the Antibodies.com catalogue, this Mouse Sp7/Osterix ELISA Kit is a sandwich enzyme-linked immunosorbent assay for the in vitro quantitative determination of mouse Sp7/Osterix in tissue homogenates, cell lysates or other biological fluids. The assay uses a microplate pre-coated with an antibody specific for Sp7/Osterix; standards and samples are added to the wells, followed by incubation with a Biotinylated Anti-Sp7/Osterix Antibody and then an HRP-Avidin conjugate. After washing, TMB substrate is added and the enzymatic reaction produces a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm in a microplate reader and the concentration of Sp7/Osterix is calculated by reference to a standard curve.

The reported assay range is 0.156–10 ng/ml with a sensitivity of <0.064 ng/ml, read colourimetrically on a microplate reader, with a total assay time of 4 hours 30 minutes. Manufacturer-reported recovery values give a mean of 91% (range 80%–102%) for serum (n=5), 90% (range 81%–100%) for EDTA plasma (n=5), and 84% (range 80%–89%) for heparin plasma (n=5). Kit components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
