From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of mouse Osteonectin (also known as SPARC) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The 96-well microtiter plate is pre-coated with a SPARC-specific capture antibody. After standards and samples are added, a Biotinylated Anti-SPARC Antibody and HRP-Avidin conjugate are used for detection. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution, and absorbance is read at 450 nm in a colorimetric microplate reader. Total assay time is 4 hours 30 minutes.

Product range is 0.312–20 ng/ml with a sensitivity of <0.144 ng/ml. Reported spike-and-recovery values (n=5): serum 80%–102% (average 91%); EDTA plasma 81%–99% (average 90%); heparin plasma 80%–89% (average 84%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16: serum 87–91%, 87–107%, 74–101% and 92–97%; EDTA plasma 90–105%, 84–101%, 90–101% and 79–108%; heparin plasma 84–95%, 92–105%, 82–105% and 89–91%.

Provided reagents include a pre-coated 96-well microplate arranged as 12 x 8 well strips, lyophilized standard, two detection solutions, wash buffer, sample dilution buffer, TMB substrate, stop solution and adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed in the protocol manual and the kit must not be used past its expiration date.
