Mouse Occludin ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com intended for the in vitro quantitative determination of mouse Occludin in tissue homogenates, cell lysates, and other biological fluids. An antibody specific for Occludin is pre-coated onto a 96-well microplate.

Standards and samples are added, where Occludin is captured by the immobilised antibody. After washing, a Biotinylated Anti-Occludin Antibody binds the captured target, followed by HRP-Avidin conjugate. TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader. The reported sensitivity is <0.117 ng/ml with a working range of 0.312–20 ng/ml. Total assay time is approximately 4 h 30 min on a pre-coated microplate arranged as 12 × 8 well strips, with a colorimetric readout.

Reported recovery in serum (n=5) spans 80–102% (average 91%); EDTA plasma (n=5) shows 81–100% (average 90%); and heparin plasma (n=5) yields 80–89% (average 84%). Dilutional linearity at 1:2, 1:4, 1:8, and 1:16 was 87–91%, 87–107%, 74–101%, and 92–97% in serum; 90–105%, 84–101%, 90–101%, and 79–108% in EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% in heparin plasma, respectively. The kit includes a pre-coated microplate, lyophilised standard, Detection Solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers, with components stored at +4°C or –20°C as instructed.
