Mouse Nuclear Factor Of Activated T-Cells, Cytoplasmic 1 ELISA Kit

A sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse Nuclear Factor Of Activated T-Cells, Cytoplasmic 1 (NFAT1) in tissue homogenates, cell lysates, or other biological fluids. The assay is reactive with mouse samples and is run on a pre-coated 96-well microplate (12 x 8 well strips), with absorbance read on a colorimetric microplate reader at 450 nm. The stated product range is 0.156–10 ng/ml and sensitivity is <0.055 ng/ml. Approximate total assay time is 4 hours 30 minutes.

The kit uses the sandwich enzyme immunoassay technique. An antibody specific for NFAT1 is pre-coated onto the wells; standards and test samples are added and NFAT1 present in each sample is bound by the immobilized antibody. After washing, a biotinylated anti-NFAT1 antibody binds the captured NFAT1, followed by HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of NFAT1 captured per well, and concentration is calculated by referring absorbance readings to the standard curve.

Spike-and-recovery performance was reported as 80–102% (average 91%) in serum (n=5), 81–101% (average 91%) in EDTA plasma (n=5), and 80–89% (average 84%) in heparin plasma (n=5). Kit components are stored at +4°C or -20°C as instructed and should not be used past the expiration date.
