Mouse Nodal Homolog ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Nodal in serum, plasma, tissue homogenates or other biological fluids. Supplied by Antibodies.com, the kit uses pre-coated microplate wells and employs the sandwich enzyme immunoassay technique. Standards and samples are added to the wells, where Nodal is captured by the immobilised antibody; following incubation and washing, a Biotinylated Anti-Nodal Antibody binds the captured target. An HRP-Avidin conjugate and TMB substrate are then used, and the resulting colour change is read at 450 nm in a microplate reader, with Nodal concentration calculated from a standard curve.

The assay has a sensitivity of <6.2 pg/ml and a quantitative range of 15.6–1000 pg/ml. Reported recovery in serum averaged 91% across a range of 80–102% (n=5), in EDTA plasma averaged 90% (range 81–100%, n=5), and in heparin plasma averaged 84% (range 80–89%, n=5). Total assay time is 4 hours 30 minutes, and kit components are stored at +4°C or -20°C as instructed; the kit should not be used past its expiration date.

Linearity has been assessed at dilutions of 1:2, 1:4, 1:8 and 1:16. In serum (n=5), observed values were 87–91%, 87–107%, 74–101% and 92–97% respectively; in EDTA plasma (n=5), 90–105%, 84–101%, 90–101% and 79–108%; and in heparin plasma (n=5), 84–95%, 92–105%, 82–105% and 89–91%.
