Mouse Nitric Oxide Synthase 2, Inducible ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay from Antibodies.com for the in vitro quantitative determination of mouse iNOS (Nitric Oxide Synthase 2, Inducible). The kit is intended for use with tissue homogenates, cell lysates, cell culture supernates and other biological fluids, with absorbance read on a colorimetric microplate reader.

The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate (12 x 8 well strips). An antibody specific for iNOS is immobilised on the plate; standards and test samples are added and any iNOS present is captured. After washing, a Biotinylated Anti-iNOS Antibody binds the captured analyte, followed by an HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow on addition of the acidic stop solution, and absorbance is read at 450 nm. Total assay time is 4h 30m.

According to the manufacturer, the measuring range is 0.312–20 ng/ml and the analytical sensitivity is <0.128 ng/ml. Reported spiked recovery values are 80–102% (average 91%, n=5) for serum, 81–100% (average 90%, n=5) for EDTA plasma and 80–89% (average 84%, n=5) for heparin plasma. Dilution linearity across 1:2 to 1:16 dilutions (n=5) is provided for the same three sample types. The kit is supplied with pre-coated plate strips, lyophilised standard, detection reagents, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or -20°C as indicated.
