From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is intended for the in vitro quantitative determination of mouse Myosin Heavy Chain 1/MYH1 in serum, plasma, tissue homogenates or other biological fluids. The assay is reactive with mouse samples and is run on a pre-coated 96-well microplate (12 x 8 well strips) format with colorimetric detection read on a microplate reader.

Quantitative measurement uses the sandwich enzyme immunoassay technique. Standards and samples are added to wells pre-coated with antibody specific for Myosin Heavy Chain 1/MYH1, followed by incubation with a Biotinylated Anti-Myosin Heavy Chain 1/MYH1 Antibody and an HRP-Avidin conjugate. The enzymatic reaction is visualised using TMB substrate solution and stopped with acidic stop solution; absorbance is read at 450 nm and the concentration calculated against the standard curve. Total assay time is 4 hours 30 minutes. Kit components include a pre-coated 96 well microplate, lyophilised standard, detection solutions A and B, wash buffer (30X), sample dilution buffer, TMB substrate, stop solution and plate sealers, to be stored at +4°C or -20°C as instructed and not used past the expiration date.

Reported performance figures include a product range of 15.6-1000 pg/ml and a sensitivity of <7.3 pg/ml. Spike-and-recovery averages were 91% in serum (range 80% - 102%, n=5), 90% in EDTA plasma (range 81% - 100%, n=5) and 84% in heparin plasma (range 80% - 89%, n=5). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 yielded 87-91%, 87-107%, 74-101% and 92-97% respectively for serum (n=5); 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma (n=5); and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma (n=5).
