Mouse Myelin Basic Protein ELISA Kit

This sandwich ELISA from Antibodies.com is designed for the in vitro quantitative determination of mouse Myelin Basic Protein in serum, plasma, tissue homogenates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, using an antibody specific for Myelin Basic Protein pre-coated onto a microplate. Captured analyte is bound by a biotinylated anti-Myelin Basic Protein antibody, followed by an HRP-avidin conjugate, with signal generated using a TMB substrate solution that produces a blue colour changing to yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the intensity of yellow colour is proportional to the amount of Myelin Basic Protein present, with concentration calculated by reference to a standard curve.

The assay has a stated sensitivity of 0.059 ng/ml and a measuring range of 0.156–10 ng/ml. Total assay time is 4 hours 30 minutes. Spike-and-recovery was evaluated with the following averages: serum (n=5) 91% (range 80%–102%), EDTA plasma (n=5) 90% (range 81%–100%), and heparin plasma (n=5) 84% (range 80%–89%). Dilution linearity was assessed at 1:2, 1:4, 1:8 and 1:16 across the same three matrices (n=5 per matrix), with observed recoveries of 87–91%, 87–107%, 74–101% and 92–97% for serum; 90–105%, 84–101%, 90–101% and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105% and 89–91% for heparin plasma.
