The assay employs a pre-coated 96-well microplate arranged as 12 x 8 well strips, onto which an antibody specific for MAdCAM1 has been immobilised. Standards and test samples are added to the wells, allowing the target analyte to be captured, followed by incubation with a Biotinylated Anti-MAdCAM1 Antibody and then an HRP-Avidin conjugate.

The reaction is visualised using TMB substrate, producing a blue product that turns yellow upon addition of acidic stop solution, with absorbance measured at 450nm in a colorimetric microplate reader. Reported sensitivity is <0.059 ng/ml with an assay range of 0.156-10 ng/ml, and total assay time is approximately 4 hours 30 minutes. Validation data show average recovery of 91% in serum (range 80-102%, n=5), 91% in EDTA plasma (range 81-101%, n=5), and 84% in heparin plasma (range 80-89%, n=5).
