Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of mouse Mitochondrial Methionyl-tRNA Formyltransferase in tissue homogenates, cell lysates or other biological fluids. The kit employs the sandwich enzyme immunoassay technique, with an antibody specific for the target pre-coated onto a 96-well microplate arranged as 12 x 8 well strips. Standards and samples are added to the wells, followed by a Biotinylated Anti-Mitochondrial Methionyl-tRNA Formyltransferase Antibody and an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and the concentration calculated from the standard curve.

The assay has a product range of 0.156–10 ng/ml and a reported sensitivity of 0.055 ng/ml. Total assay time is 4 hours 30 minutes, with detection performed on a colorimetric microplate reader. Kit components include a pre-coated 96-well microplate, lyophilized standard (2 vials), Detection Solution A (120 µl), Detection Solution B (120 µl), 30X Wash Buffer (20 ml), Sample Dilution Buffer (45 ml), TMB Substrate (9 ml), Stop Solution (6 ml), and plate sealers (5 adhesive strips). Components should be stored at +4°C or -20°C as instructed and not used past the expiration date.

Recovery has been validated across sample matrices: serum (range 80%–102%, average 91%, n=5), EDTA plasma (range 81%–100%, average 90%, n=5), and heparin plasma (range 80%–89%, average 84%, n=5). Dilutional linearity data are provided for serum, EDTA plasma, and heparin plasma across 1:2, 1:4, 1:8, and 1:16 dilutions. The kit is suitable for cell lysates and tissue homogenates.
