Mouse Microtubule Associated Protein Tau ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com designed for the in vitro quantitative determination of mouse Microtubule Associated Protein Tau in tissue homogenates, cell lysates, or other biological fluids.

The assay uses a pre-coated 96-well microtiter plate (12 x 8 well strips). Standards and samples are added to the wells, where the target binds the immobilized capture antibody. Following incubation and washing, a Biotinylated Anti-Tau Antibody is added, followed by HRP-Avidin conjugate. TMB substrate produces a blue-coloured product that turns yellow after addition of acidic stop solution. Absorbance is read at 450 nm using a colorimetric microplate reader, and the target concentration is calculated from the standard curve. The assay has a reported range of 7.81–500 pg/ml and a sensitivity of <2.9 pg/ml, with a total assay time of 4 hours 30 minutes.

Recovery validation (n=5) showed ranges of 80%–102% (average 91%) in serum, 81%–100% (average 90%) in EDTA plasma, and 80%–89% (average 84%) in heparin plasma. Linearity of dilution (n=5) at 1:2, 1:4, 1:8, and 1:16 produced recoveries of 87–91%, 87–107%, 74–101%, and 92–97% in serum; 90–105%, 84–101%, 90–101%, and 79–108% in EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% in heparin plasma. Kit components are stored at +4°C or -20°C as instructed, and the kit should not be used past the expiration date.
