Description
This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse Microtubule Associated Protein 2 (MAP2) in serum, plasma, tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for MAP2 pre-coated onto a 96-well microtiter plate. Standards and samples are added to the wells, where MAP2 is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-MAP2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution produces a blue product that changes to yellow upon addition of acidic stop solution; the density of yellow is proportional to the amount of MAP2 captured and is read at 450 nm in a microplate reader.
The kit has an assay range of 0.156–10 ng/ml, a sensitivity of 0.053 ng/ml, and a total assay time of 4h 30m. The platform is a pre-coated microplate comprising 12 x 8 well strips, intended for use with a colorimetric microplate reader. Recovery in serum (n=5) ranges from 80% to 102% with an average of 91%; in EDTA plasma (n=5), 81% to 100% with an average of 90%; and in heparin plasma (n=5), 80% to 89% with an average of 84%. Linearity has been demonstrated across dilutions of 1:2, 1:4, 1:8 and 1:16 in the same three sample types.
Kit components should be stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date.
