Mouse Methyl CpG Binding Protein 2 ELISA Kit

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse MeCP2 in tissue homogenates, cell lysates, or other biological fluids. The kit uses a 96-well microplate pre-coated with an antibody specific for MeCP2. Standards and samples are added to the wells, where MeCP2 is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-MeCP2 Antibody binds the captured analyte, then HRP-Avidin conjugate is added. TMB substrate is used to visualise the enzymatic reaction, producing a blue product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of MeCP2 captured, and the concentration is calculated by reading the O.D. absorbance at 450nm in a microplate reader against a standard curve.

The assay has a sensitivity of 0.102 ng/ml with a range of 0.312-20 ng/ml. Reported average recovery values are 91% for serum (range 80-102%), 90% for EDTA plasma (range 81-100%), and 84% for heparin plasma (range 80-89%). Total assay time is approximately 4 hours 30 minutes. The kit includes a pre-coated 96-well microplate arranged as 12 x 8 well strips, lyophilised standard, Detection Solution A and B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and adhesive plate sealers. Components are stored at +4°C or -20°C as instructed, and detection is colourimetric using a standard microplate reader.
