Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse Matrix Metalloproteinase 14 (MMP14) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique: an antibody specific for MMP14 is pre-coated onto a microplate, standards and samples are added, and captured MMP14 is detected with a Biotinylated Anti-MMP14 Antibody followed by an HRP-Avidin conjugate. A TMB substrate produces a blue product that turns yellow upon addition of acidic stop solution; absorbance is read at 450 nm in a microplate reader and MMP14 concentration is calculated against the standard curve. The detection method is colorimetric and the assay is quantitative.

Reported performance figures include a sensitivity of 32.81 pg/ml and an assay range of 93.8–6000 pg/ml, with the standard curve prepared as a 1:2 dilution series from 6000 pg/ml down to 93.8 pg/ml. Total assay time is approximately 4 hours 30 minutes. Sample recovery values (n=5) are reported as 83%–100% with an average of 91% for serum, 90%–103% with an average of 96% for EDTA plasma, and 89%–102% with an average of 95% for heparin plasma. Dilution linearity (n=5) for serum is 83–110% at 1:2, 79–100% at 1:4, 90–98% at 1:8 and 82–108% at 1:16; for EDTA plasma 90–98%, 90–105%, 73–109% and 87–91% across the same dilutions; and for heparin plasma 88–103%, 77–105%, 95–98% and 87–110%.

Kit components include a pre-coated microplate, lyophilized standard, Detection Solution A and Detection Solution B, 30X Wash Buffer, Sample Dilution Buffer, TMB Substrate, Stop Solution and plate sealers, which are stored at +4°C or -20°C as instructed. The product is for research use only and is not intended for diagnostic or therapeutic use.
