Mouse Matrix Gla Protein ELISA Kit

This Antibodies.com sandwich ELISA kit enables the in vitro quantitative determination of mouse Matrix Gla Protein (Mgp) in serum, plasma, tissue homogenates and other biological fluids. The 96-well microplate is pre-coated with an antibody specific for Mgp; standards and samples are added and the target is captured by the immobilised antibody. After washing, a biotinylated anti-Mgp antibody binds the captured analyte and an HRP-avidin conjugate is then added. TMB substrate is used to visualise the HRP reaction, producing a blue colour that turns yellow upon addition of stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader and the Mgp concentration is calculated from the standard curve.

Reported sensitivity is <33 pg/ml with a measurable range of 78.1-5000 pg/ml, and total assay time is approximately 4 hours 30 minutes. Spike-and-recovery testing gave average recoveries of 91% in serum (n=5, range 80-102%), 90% in EDTA plasma (n=5, range 81-100%) and 84% in heparin plasma (n=5, range 80-89%). Dilution linearity at 1:2, 1:4, 1:8 and 1:16 respectively was 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma.
