From Antibodies.com, this Mouse LBP ELISA Kit is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse Lipopolysaccharide Binding Protein (LBP) in serum, plasma or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, with an antibody specific for LBP pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells and the LBP present in each sample is bound to the wells by the immobilized antibody.

Following incubation and washing, the wells are incubated with a Biotinylated Anti-LBP Antibody, which binds the captured LBP, followed by addition of an HRP-Avidin conjugate. TMB substrate solution is used to visualize the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow after addition of acidic stop solution. The density of yellow is proportional to the amount of LBP captured in each well, and the concentration of LBP is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve. The assay time is 4 hours 30 minutes.

The kit has a product range of 1.56-100 ng/ml and a sensitivity of <0.61 ng/ml. Reported recovery averages across 5 samples per type are 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%), and 84% for heparin plasma (range 80%-89%). Linearity has been validated at dilutions of 1:2, 1:4, 1:8 and 1:16 across serum, EDTA plasma and heparin plasma. The pre-coated microplate is supplied as 12 x 8 well strips, and kit components are stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date.
