Mouse Linker For Activation Of T-Cell ELISA Kit

This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com enables the in vitro quantitative determination of mouse LAT (Linker For Activation of T-Cell) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids. The kit employs a 96-well microtiter plate pre-coated with an antibody specific for LAT.

Standards and test samples are added to the wells, where captured LAT is detected using a biotinylated anti-LAT antibody, followed by an HRP-avidin conjugate and TMB substrate. The enzymatic reaction produces a blue-coloured product that turns yellow upon addition of acidic stop solution; absorbance is then read at 450 nm on a colorimetric microplate reader and the LAT concentration is calculated from the standard curve. The quantitative assay has a working range of 0.156–10 ng/ml and a sensitivity of 0.039 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery has been characterised in serum (80%–102%, average 91%, n=5), EDTA plasma (81%–100%, average 90%, n=5) and heparin plasma (80%–89%, average 84%, n=5).

Dilution linearity has been validated across 1:2, 1:4, 1:8 and 1:16 dilutions for serum, EDTA plasma and heparin plasma samples. Kit components are stored at +4°C or -20°C as indicated, and the kit should not be used past its expiration date.
