Description

A sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com, designed for the in vitro quantitative determination of mouse Leukocyte Cell Derived Chemotaxin 2 (LECT2) in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Reactivity is mouse, the platform is a pre-coated microplate (12 x 8 well strips), detection is colourimetric, and total assay time is 4 hours 30 minutes.

The kit employs a sandwich enzyme immunoassay technique in which an antibody specific for LECT2 is pre-coated onto a 96-well microtiter plate. Standards and test samples are added to the wells and LECT2 present in each sample is captured by the immobilised antibody. Following incubation and washing, a Biotinylated Anti-LECT2 Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of LECT2 captured in each well, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader and referring to the standard curve.

The product range is 31.2–2000 pg/ml with a sensitivity of <13.6 pg/ml. Reported average recovery values from spiked samples (n=5) are: serum 80%–102% (average 91%), EDTA plasma 81%–100% (average 90%), and heparin plasma 80%–89% (average 84%). Dilution linearity for serum, EDTA plasma and heparin plasma (n=5 each) at 1:2, 1:4, 1:8 and 1:16 dilations is provided in the package insert. Kit components are to be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date.
