Mouse Klotho ELISA Kit

From Antibodies.com, this sandwich ELISA kit is designed for the in vitro quantitative determination of mouse Klotho in serum, plasma, tissue homogenates, or other biological fluids. The quantitative colorimetric assay uses a pre-coated 96-well microplate in a 12 x 8 well strip format. A biotinylated anti-Klotho detection antibody is added after sample binding, followed by HRP-Avidin conjugate. The enzymatic reaction is visualised with TMB substrate, producing a blue colour that turns yellow upon addition of acidic stop solution, and the absorbance is read at 450 nm on a colorimetric microplate reader. Total assay time is 4 hours 30 minutes.

The kit reports a sensitivity of <5.1 pg/ml and an assay range of 12.5–800 pg/ml. Spike-and-recovery values are reported at averages of 91% for serum (n=5, range 80–102%), 90% for EDTA plasma (n=5, range 81–99%), and 84% for heparin plasma (n=5, range 80–89%). Linearity upon dilution at 1:2, 1:4, 1:8, and 1:16 is given as 87–91%, 87–107%, 74–101%, and 92–97% for serum; 90–105%, 84–101%, 90–101%, and 79–108% for EDTA plasma; and 84–95%, 92–105%, 82–105%, and 89–91% for heparin plasma.
