Mouse Keratin 18 ELISA Kit

This Sandwich ELISA kit is designed for the in vitro quantitative determination of mouse Cytokeratin 18 in serum, plasma, tissue homogenates, cell lysates, cell culture supernates or other biological fluids. Reactivity is Mouse, with Cytokeratin 18 (Keratin 18) as the protein target.

The assay employs the sandwich enzyme immunoassay technique on a pre-coated 96-well microplate (12 x 8 well strips). An antibody specific for Cytokeratin 18 is immobilised on the plate; standards and samples are added, followed by a Biotinylated Anti-Cytokeratin 18 Antibody and an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow after addition of stop solution. The density of yellow is proportional to the amount of Cytokeratin 18 captured, read as O.D. absorbance at 450 nm on a colorimetric microplate reader. The product range is 78.1-5000 pg/ml, with a sensitivity of 25 pg/ml. Assay time is 4h 30m, and kit components are stored at +4°C or -20°C as instructed.

Reported recovery values are: Serum (n=5) 83-100%, average 91%; EDTA Plasma (n=5) 90-103%, average 96%; Heparin Plasma (n=5) 89-102%, average 95%. Linearity upon dilution at 1:2, 1:4, 1:8 and 1:16 is given as: Serum 83-110%, 79-100%, 90-98%, 82-108%; EDTA Plasma 90-98%, 90-105%, 73-109%, 87-91%; Heparin Plasma 88-103%, 77-105%, 95-98%, 87-110%.
