Offered by Antibodies.com, this is a sandwich Enzyme-Linked Immunosorbent Assay (sELISA) designed for the in vitro quantitative determination of mouse JunB in tissue homogenates or other biological fluids. The assay employs the sandwich enzyme immunoassay technique using an antibody specific for JunB pre-coated onto a microplate. Standards and test samples are added to the wells and the JunB present in each sample is bound by the immobilized antibody. After incubation and washing, a Biotinylated Anti-JunB Antibody is added, followed by HRP-Avidin conjugate. TMB substrate solution is then used to visualise the HRP enzymatic reaction, producing a blue-coloured product that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of JunB captured, and the concentration is calculated by reading the O.D. absorbance at 450 nm in a microplate reader.

The kit has a reported sensitivity of <0.042 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4 hours 30 minutes. Recovery data for n=5 samples shows averages of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Dilution linearity is reported at 1:2, 1:4, 1:8, and 1:16 for serum, EDTA plasma, and heparin plasma.

Kit components include a pre-coated microplate, lyophilised standard, Detection Solution A, Detection Solution B, Wash Buffer (30X), Sample Dilution Buffer, TMB Substrate, Stop Solution, and plate sealers. Components are stored at +4°C or -20°C as instructed, and the kit should not be used past its expiration date. The assay is read using a colorimetric microplate reader.
