This sandwich Enzyme-Linked Immunosorbent Assay (sELISA) from Antibodies.com is intended for the in vitro quantitative determination of mouse Islet Amyloid Polypeptide, also referred to in the product literature as Amylin/DAP. The kit is reactive with mouse samples and is validated for use with serum, plasma, tissue homogenates, cell lysates, cell culture supernates and other biological fluids.
The assay is run on a pre-coated 96-well microplate (12 x 8 well strips) and read on a colorimetric microplate reader at 450 nm. It has a stated product range of 78.1–5000 pg/ml and a sensitivity of <24 pg/ml, with a total assay time of 4h 30m. Kit components should be stored at +4°C or −20°C as instructed, and the kit should not be used past its expiration date. Components supplied include the pre-coated plate, lyophilized standard, detection solutions A and B, wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers.
Recovery performance reported for serum (n=5) was 80–102% (average 91%), for EDTA plasma (n=5) was 81–100% (average 90%), and for heparin plasma (n=5) was 80–89% (average 84%). Across 1:2, 1:4, 1:8 and 1:16 dilutions, recoveries ranged 74–107% for serum (n=5), 79–108% for EDTA plasma (n=5) and 82–105% for heparin plasma (n=5).
