Mouse Involucrin ELISA Kit

This sandwich enzyme-linked immunosorbent assay (sELISA) from Antibodies.com is designed for the in vitro quantitative determination of mouse Involucrin in serum, plasma, tissue homogenates and other biological fluids.

The assay employs a pre-coated 96-well microplate (12 x 8 well strips) onto which an antibody specific for Involucrin has been immobilised. Standards and samples are pipetted into the wells, where Involucrin is captured by the immobilised antibody. After incubation and washing, a Biotinylated Anti-Involucrin Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the reaction, producing a blue product that turns yellow upon addition of the stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and concentration is calculated by reference to the standard curve.

Key performance figures include a sensitivity of <0.067 ng/ml and an assay range of 0.156–10 ng/ml, with a total assay time of 4h 30m. Spiking recovery in serum (n=5) averaged 91% (range 80–102%), in EDTA plasma (n=5) averaged 90% (range 81–100%), and in heparin plasma (n=5) averaged 84% (range 80–89%). Dilution linearity was demonstrated at 1:2, 1:4, 1:8 and 1:16 dilutions across serum, EDTA plasma, and heparin plasma. Kit components include a pre-coated microplate, lyophilised standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution and plate sealers, stored at +4°C or −20°C as instructed.
