Description

From Antibodies.com, this sandwich Enzyme-Linked Immunosorbent Assay (sELISA) is designed for the in vitro quantitative determination of mouse CXCR2 — also referred to as Interleukin 8 Receptor Beta — in serum, plasma, tissue homogenates, cell lysates or other biological fluids.

The kit employs the sandwich enzyme immunoassay technique. An antibody specific for CXCR2 is pre-coated onto a 96-well microplate supplied as 12 x 8 well strips. Standards and test samples are added to the wells; after incubation and washing, a Biotinylated Anti-CXCR2 Antibody is added, followed by an HRP-Avidin conjugate. A TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow on addition of the acidic stop solution. The density of yellow is proportional to the amount of CXCR2 captured, and concentration is calculated from O.D. absorbance at 450 nm against a standard curve. Detection is colourmetric. The assay range is 31.2-2000 pg/ml with a sensitivity of <12.4 pg/ml, and total assay time is 4h 30m.

Reported recovery (average, n=5) is 91% for serum (range 80%-102%), 90% for EDTA plasma (range 81%-100%) and 84% for heparin plasma (range 80%-89%). Linearity across dilutions of 1:2, 1:4, 1:8 and 1:16 is 87-91%, 87-107%, 74-101% and 92-97% for serum; 90-105%, 84-101%, 90-101% and 79-108% for EDTA plasma; and 84-95%, 92-105%, 82-105% and 89-91% for heparin plasma. Kit components are a Pre-Coated 96 Well Microplate (12 x 8 well strips, -20°C), Lyophilised Standard (2 vials, -20°C), Detection Solution A (120 μl, -20°C), Detection Solution B (120 μl, -20°C), 30X Wash Buffer (20 ml, +4°C), Sample Dilution Buffer (45 ml, -20°C), TMB Substrate (9 ml, +4°C), Stop Solution (6 ml, +4°C) and 5 adhesive plate sealers. Components should be stored at +4°C or -20°C as instructed and not used past the expiration date.
