A sandwich ELISA designed for the in vitro quantitative determination of mouse CXCR1 in tissue homogenates, cell lysates, and other biological fluids. The assay is supplied by Antibodies.com and is reactive with mouse samples, targeting CXCR1 (also known as Interleukin 8 Receptor Alpha). The assay is built around a pre-coated 96-well microplate arranged as 12 × 8 well strips, with a CXCR1-specific capture antibody immobilised on the plate surface.

Standards and test samples are added to the wells, where CXCR1 is captured by the immobilised antibody. After washing, a biotinylated anti-CXCR1 detection antibody is added, followed by an HRP-avidin conjugate. TMB substrate is then used to visualise the enzymatic reaction, producing a blue product that turns yellow upon addition of an acidic stop solution. Absorbance is read at 450 nm on a colorimetric microplate reader, and the concentration of CXCR1 is calculated against the standard curve. The full protocol takes 4 hours 30 minutes. Reported sensitivity is <0.054 ng/ml and the working assay range is 0.156–10 ng/ml.

In spiked recovery testing (n=5 per matrix), the manufacturer reports average recoveries of 91% for serum (range 80–102%), 90% for EDTA plasma (range 81–100%), and 84% for heparin plasma (range 80–89%). Kit components should be stored at +4°C or –20°C as indicated, and the kit should not be used past its expiration date.
