A sandwich Enzyme-Linked Immunosorbent Assay (ELISA) from Antibodies.com, designed for the in vitro quantitative determination of mouse Interleukin 6 Receptor (IL-6R) in serum, plasma, tissue homogenates or other biological fluids. The kit is reactive with mouse samples and uses a pre-coated 96-well microplate (12 x 8 well strips) read colourimetrically on a microplate reader.

The assay employs the sandwich enzyme immunoassay technique. An antibody specific for IL-6R is pre-coated onto the plate; standards and test samples are added and IL-6R is captured by the immobilized antibody. After washing, a Biotinylated Anti-IL-6R Antibody is added, followed by an HRP-Avidin conjugate. TMB substrate is then used to visualise the HRP enzymatic reaction, producing a blue product that turns yellow upon addition of acidic stop solution. Absorbance is read at 450 nm and concentration is calculated against the standard curve. Reported total assay time is 4h 30m.

Stated performance figures include an assay range of 31.2-2000 pg/ml and a sensitivity of <11.4 pg/ml. Reported sample recovery across dilutions (1:2, 1:4, 1:8, 1:16) includes serum (n=5) at 81-99%, 90-102%, 77-104% and 88-95%; EDTA plasma (n=5) at 94-104%, 80-108%, 88-93% and 86-108%; and heparin plasma (n=5) at 76-100%, 92-99%, 88-105% and 85-99%. Kit components include lyophilised standard (2 vials), Detection Solution A (120μl), Detection Solution B (120µl), Wash Buffer 30X (20ml), Sample Dilution Buffer (45ml), TMB Substrate (9ml), Stop Solution (6ml) and 5 adhesive plate sealers, with components stored at +4°C or -20°C as instructed. The kit should not be used past its expiration date.
