This sandwich ELISA kit from Antibodies.com is designed for the in vitro quantitative determination of mouse IL-5RA in serum, plasma, or other biological fluids. The assay employs the sandwich enzyme immunoassay technique, using an antibody specific for IL-5RA that has been pre-coated onto a 96-well microtiter plate.

In the workflow, standards and test samples are added to the wells and IL-5RA present in each sample is bound by the immobilized antibody. After washing, a Biotinylated Anti-IL-5RA Antibody is added, followed by an HRP-Avidin conjugate. Following further incubation and washing, TMB substrate solution is used to visualize the enzymatic reaction, producing a blue colour that changes to yellow upon addition of acidic stop solution. The density of yellow is proportional to the amount of IL-5RA captured, and concentration is calculated by reading absorbance at 450 nm in a colorimetric microplate reader. The assay has a sensitivity of <28.5 pg/ml and a range of 62.5–4000 pg/ml, with a typical total assay time of 4h 30m on a pre-coated microplate platform of 12 x 8 well strips.

Reported recovery data shows averages of 91% for serum (n=5), 90% for EDTA plasma (n=5), and 84% for heparin plasma (n=5). Kit components include a pre-coated 96 well microplate (12 x 8 well strips), lyophilized standard, detection solutions A and B, 30X wash buffer, sample dilution buffer, TMB substrate, stop solution, and plate sealers. Storage is at +4°C or −20°C depending on the component, and the kit should not be used past its expiration date.
